I actually asked Trick Dempsey on the origins of Hulkers during the friday livestream (Myll_Eric helped me convey the question to him), sad thing I can't remember even half of the answer (was a bit tired, the defiance portion was in the middle of the night for me, so I was pretty tired). But hulkers are basically mutants anyway, lol.
The Mutant "faction" just consists of the EMC guys who got mutated. I don't think that in any way precludes there being other mutants in the world.
Hulkers are biomen, mutated to their maximum "potential."

Hehe, I saw the commercials and thought the game would be dumb after they showed Hulkers. It just looked lame on the TV ad, like they were spoiling the realism of the game by adding huge men with exposed brains under their helmets. But in game, it works quite well. I solo the Wandering Hulker emergency every time I find it.
I'm sure we'll see one of these characters in the TV series before too long... although why they are teamed up with raiders is beyond me.
