I believe you have to point it to the /live subdirectory, there are a few other threads about it you might want to check.

^ Yes, ty, I had tried that but did not work.
This finally worked; closed Glyph, then moved/dragged all files/subs out of /Live down into root /Defiance. Deleted empty /Live, also deleted files "Patcher" and "Manifest".
Ran Glyph, clicked Patch, this time it re-checked the game files for a few minutes and finally confirmed they were all valid.
Please improve Patcher AI !.
