It happened to me and another player yesterday. I ran over to the ammo crate and logged out. When I logged back in and ran towards the spot, the captured ranchers were there. I was able to advance and complete the mission.
Start a competitive multiplayer match. That's what I did, and when I returned to the quest the ranchers were there.
i know how you feel my friend i have done the quest about 20-30 times now and the rancher in the building has got stuck every time

The mission is still bugged. I think it's because the ranger keeps running back and forth instead of running into the correct destination.
Spent most of my evening trying to get past this mission, no dice (no ranchers either) just me and about 15-20 other people killing raider spawns, logging, relogging, abandoning & restarting doing some other missions/coop etc... and still no ranchers. Waiting for a server reset to see if that'll sort matters but kind of bad for a main story mission.

Myself and at least 40 other players are also suffering the same glitch, on Xbox 360, 4/9/2013, glitch persisted from 6pm EST through 8pm EST, and probably still persists now. I quit trying to complete it after reading the numerous glitch threads.
At the first waypoint, in the field where there is a large Mower, kinda looks like a bulldozer and a truck that raiders spawn out of. The raiders spawn, but never any NPCs to rescue.
Love the game so far, and hope you get these issues fixed quickly so my less patient friends can play with me!
I experienced this bug, but then I got past it on a second attempt. However, upon reaching Miwok Ranch, there's yet another bug where the game won't allow you to free the last captured rancher. I'm finished playing until it's fixed. Not much else to do.
