If the game and TV are seperated by over 2000 miles then how can they be linked together in any way? Its a post appocalyptic world and media is only localised at the most due to lack of satalites.
So how would the people in Defiance or why would they care what happens over 2000 miles away ?
You do realize that world travel has been possible without tremendous technology, and as far as that is there still is aerial vehicles so travel wouldnt be that big of a problem, and aliens, I mean cmon they gotta know how to travel across a planet or communicate considering they came from somewhere other than earth in spaceships... lol
Personally I think that eventually Defiance will become the main hub in-game, once the show progresses far enough.

Personally I would much rather have a chance to visit Antartica if nothing more for a short visit, though I belive the people saying "it will never happen" could be wrong.
If/when the show one day ends, depending on the ending if there is one, It would make perfect sense to use the city of defiance for a expansion since it already comes with a rich enviroment and chars that players would know and and love from the series.
However as long as the show is ongoing, I don't see it ever happening.
Seems kinda weird to call the game Defiance if you never go there, but I guess we'll see. Also why does real geography actually matter? it's not like you will physically have to travel across the continent. They could just place a plane/carrier and just be like click this, bam you're in Defiance
There's not really any massive reason that we couldn't progress to Defiance even whilst the show is running. So that's where the show is set. big deal it's not like we'd be killing off main characters.
Defiance was a point in time, when we fought back. It spawned a battle, quite obviously. Later a place was named after it.
Pretty sure it says exactly that in media for either the game or the show but I don't recal which.
