I'v only heard that the characters from the game (NPC) will be on the show, while they leave game. So they won't be found in game because they are in the show (both show and game are in separate areas)

Watch the Comiccon 2012 defiance interview video..its mentioned nearer the end of the interview by the game devs & tv show producer.
The video is pretty good as it mentions a lot of ideas that they want to do and includes a Q&A with the cast.

I always was under the impression that it was more along the lines of Guilds having a chance to being mentioned by name by characters during the show.
And NPCs moving back and forth between the game and the show.
Angry Joe had a dev-interview where it was said that if a guild does really well they'll get "a mention" on the show and that there's already "blanks in place" (sort of like Mad Libs) for cross-over and such.
