Gears of War, seems to be most like what Defiance is like with the 3rd person aspect.

Even though its an older game, if you can play any 3rd person shooters like Socom(which is on PS) or Crackdown, it might help you get used to playing 3rd person shooters. It can be difficult to make the transition from a 1st person to a 3rd person for some people.

Mass effect 3 look a lot like the game play of defiance but slower pace

I'd recommend Halo 4. It's not a 3rd person shooter but it should teach you to be a decent shot. It also has vehicles similar to the quad bike and jeep i've seen in Defiance screenshots.
